Measure! I am currently building a Size 5 from the Antes plans. With a scale length of 21.375" using StewMac's fret calculator, it gives me a high E string length of 21.450" and a low E string length of 21.555" with compensation. The bridge plate that I am using will be only slightly wider than the bridge and if I had just followed the plans it would have been out of position by about 3/16". Not sure if it's an error in the plans or just a printing error, but don't forget to MEASURE!

The Antes plans don't include any compensation at all in the layout?

Absolutely...!! When Laying out the brace pattern on the top, measure and mark where the bridgeplate and front edge of the saddle will fall. Transfer the edge of the saddle onto the bridgeplate after gluing. Then when marking for routing the linings for braces on the top, make sure your saddle mark is in the right position relevant to the fret located at the top edge of the body.

When I made my Size5 from the Antes plans, I used a 21.308" scale length. That way I was able to use my StewMac 25.34" Fret Template starting at the 3rd fret mark as the nut. Made cutting the fret slots pretty easy, not having to make a new template.
